Messenger Bio is happy to offer a range of Cy5 labeled mRNA products for your research. In fact, we offer Cy5-labeling on all our off-the-shelf and custom mRNAs. Our high-quality mRNAs are synthesized with Cyanine 5-conjugated Uridine Triphosphate (Cy5-UTP) in a 1:3–1:5 ratio (Cy5-UTP:UTP), providing you with a powerful tool for imaging and tracking mRNA localization independent of the translation stage. Our Cy5-labeled mRNA can also be used for nanoparticle loading quantification assays.

All Cy5-labeled mRNAs include a Cap1 structure (biologically superior to Cap0), a highly expressing 5’ UTR and highly stable 3’ UTR, a 100-nt poly(A) tail, and a codon-optimized open reading frame. Choose from unmodified or modified (N1-methylpseudouridine or 5-methoxyuridine) bases. Both types of modified bases have been shown to enhance protein expression in vitro and in vivo.

We offer Cy5 labeling on all our mRNA products, ensuring a perfect match for your specific research requirements. Cy5-UTP emits a bright red fluorescence upon excitation using the 647 channel, allowing for easy and accurate detection of intact mRNA molecules. It’s also suitable for a wide range of applications, including fluorescence in situ hybridization (FISH), microarray analysis, and other nucleic acid-based assays.

All Cy5-labeled mRNAs come ready-to-use in a 1 mM sodium citrate buffer (pH 6.5), which is ideal for preserving RNA stability and extending shelf-life. Just complex with your favorite transfection reagent and you're ready to go. Alternatively, speak to us about our ready-to-transfect LNP formulations.

Important Note: transfection with Cy5-labeled mRNA will result in somewhat lower mRNA translation and protein expression compared to unlabeled mRNA, due to the presence of the Cy5 moiety in the mRNA strand. We can reduce the extent to which translation is impacted by reducing the Cy5-UTP:UTP ratio and/or by substituting UTP for M1ψTP or 5moUTP.
